This medium is suitable for various types of mammalian cells, including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. RPMI 1640 contains a glutathione reducer and high concentrations of vitamins, some of which are not present in Eagle’s Minimal Essential Medium or Dulbecco’s Modified Eagle Medium.

This medium is deprived of proteins, lipids, or growth factors, so it is usually supplemented with 10% fetal bovine serum (FBS). The sodium bicarbonate buffer system allows for maintaining physiological pH in an environment with 5-10% CO2 gas.

Researchers can choose from various modifications of RPMI 1640 based on specific needs, such as glutamine supplementation, phenol red, and other additives.
