Loading buffers are used to prepare and visualize DNA-containing samples for electrophoresis on agarose or polyacrylamide gels. After mixing each of the loading buffers with the sample, its density and compactness cause the sample to become heavy. Therefore, the sample is placed at the bottom of the gel wells, preventing the sample from spreading in the electrophoresis buffer. These buffers also inhibit the activity of many nucleases. Additionally, the observation of DNA bands will not be problematic due to the presence of the residual effect of this color. This feature is very important in performing the Ladder electrophoresis. Bromophenol blue, the standard color present in this buffer, is used for visual tracking of DNA movement during electrophoresis. The movement of bromophenol blue is approximately 300 base pairs in a 1% agarose and TBE standard gel.
