MLPA is a semi-quantitative and non-automated technique used to determine the relative copy number of up to 60 DNA sequences in a single reaction, based on multiplex PCR.<\/p>\n<p>The principle of MLPA involves amplifying and replicating approximately 60 probe sequences, each targeting specific DNA sequences of around 60 base pairs in length.<\/p>\n<p>Figure 1: MLPA reaction results in the production of a set of unique PCR amplicons ranging in length from 64 to 500 base pairs, which are separated by capillary electrophoresis. After initial denaturation of the DNA template, a mixture of MLPA probes is added to it. As a result, a set of unique PCR amplicons is created. One PCR primer is fluorescently labeled, which allows the amplified PCR products to be visualized during separation in the genetic analyzer device.
